An Integrated Approach to Accelerate the ADC Discovery with WuXi XDC Innovative Technology Toolbox

Discovery efforts for antibody-drug conjugates (ADCs) and other bioconjugates require broad expertise across multiple disciplines, including antibody discovery, linker-payload design and synthesis, subsequent conjugation, and in vitro and in vivo characterization studies. Scientists constantly face numerous challenges throughout the discovery process, such as fragmented CRO networks, challenging conjugation processes, analytical characterization of diversified moieties, and access to innovative technologies. Among these challenges, utilizing the right technology to precisely control the drug-to-antibody ratio (DAR) and drug distribution, as well as developing stable linkers to overcome ADC instability issues, are common in the ADC field.

 

WuXi XDC has built an integrated bioconjugate discovery platform that includes antibody development, linker payload design, ADC/bioconjugate preparation, developability assessment, proprietary narrow DAR distribution technology, stable linker technology, and in vitro/in vivo characterization. The DAR is one of the most critical quality attributes for ADCs. WuXi XDC has developed various DAR analytical methods tailored to different molecules. The WuXiDARx technology can significantly improve ADC homogeneity for desired specific DAR species such as DAR2, DAR4, and DAR6. The X-LinC technology can significantly improve the plasma stability often seen problems with maleimide linkers. The integrated ADC discovery approach, combined with WuXi XDC’s innovative technologies and developability assessment toolbox, enables seamless alignment and accelerates the ADC and bioconjugate drug discovery process.
