As an innovative bioconjugate modality combining antibody targeting and oligonucleotide-mediated gene modulation, AOC faces manufacturability challenges especially poor yields when scaling site-specific DAR2 batches for preclinical use.

 

This case features the non-GMP pilot material manufacturing of site-specific DAR2 Antibody-Oligonucleotide Conjugate (AOC) based through engineered cysteine conjugation from WuXi XDC.
 
Leveraging comprehensive process optimization, the project successfully scales up production to 100g batch scale at non-GMP pilot plant, the overall process yield is dramatically elevated from original 40% to over 70%, resolving the core yield bottleneck in AOC scale-up.
