Application of Advanced Technologies in the Manufacturing of Payload-Linker Products

This poster presents 3 advanced technologies that address key challenges in payload linker (PL) production. 1) Flow hydrogenation that offers a safer and more efficient alternative to traditional batch-mode hydrogenation, enabling high conversion rates while minimizing risks in high-potency environments. 2) A post-column enrichment strategy that significantly reduces the volume of prep-HPLC fractions—by over 75%—prior to lyophilization. This approach shortens processing time and mitigates decomposition risks associated with residual acidic modifiers, preserving payload integrity. 3) The identification and control of conjugatable impurities based on mock-conjugation derivatization and LC-MS, to characterize and manage these impurities early in payload-linker development. These innovations collectively enhance safety, efficiency, and product quality, supporting the robust manufacture of payload-linkers in large quantity for next-generation ADCs.
