Dual-Payload ADCs by Design: Integrating WuXiDARx™ Conjugation with Hydrophilic WuXi Payload-Linkers™
Dual-payload antibody-drug conjugates (dp-ADCs) represent an emerging ADC format with the potential to address tumor heterogeneity and payload resistance. By combining payloads with complementary or orthogonal mechanisms of action, dp-ADCs may enhance antitumor activity and overcome resistance. However, the dp-ADC development requires precise control of payload loading and ratio while managing the hydrophobicity and structural complexity associated with incorporating two payloads.
In this scientific poster, we present how WuXiDARx™ and WuXi Payload-Linker™ technologies enable the rational design and streamlined development of dual-payload ADCs with defined payload ratios, improved physicochemical properties, and enhanced antitumor activity.
