The next generation of ADCs are evolving through parallel innovations in target engagement and payload-linker innovations, dominated by bispecific ADC (BsADC) and dual-payload ADC(dp-ADC). Due to the increased complexity of such modality, these programs are calling for precise conjugation strategies, optimized payload-linker architectures, and improved control of physicochemical properties. The WuXiDARx Conjugation platform offers a flexible target DAR, flexible DAR choices for dual-payload ADC discovery, with a simple and scalable conjugation process. WuXiTecan2, a payload linker engineered to mitigate exatecan-associated hydrophobicity, enabled dual-payload ADC screening paired with TOP1i with favorable chemical properties.
